The locked door is inside that tunnel you're trying to get in. You need to go around the far side of the tunnel where the door will let you in, when you go down and deal with the rats, halfway along on the left there is a locked door which your key opens.

If you go to the lower section of the settlement, prepare to deal with a couple Starved Hounds, an Evangelist and a few Peasant Hollows. Note the entrance underneath the stairs on your right. This leads to a sewer, a number of optional underground pathways and an alternate path to the Dilapidated Bridge bonfire.

This is Loretta, but you need to unceremoniously cut her down, then go down through the house and out in order to actually collect the bone.

Get on that, and go straight up the stairs to find the giant, who is remarkably friendly and will agree to stop throwing his (seemingly inexhaustible supply of) spears at you.

From the tower with the giant, get on the lift back down and listen out for Siegward of Carim doing his lost-in-thought “Hmmm” noise, because around about there you should see a platform that you can jump to from the lift.
